The planet killer destroyed the planets in system L-374, including a planet where the crew of the Constellation had taken refuge. Enterprise personnal programmend the hulk of the Constellation to self-destruct its impulse drive inside the planet killer, destroying the robotic weapon.
TOS "The Doomsday Machine"
The miniature Constellation, identical in design to the original E n t e r p r i s e, was an AMT plastic Enterprise model kit, appropriately burnt and scorched. The Starship Constellation was presumably replaced by a starship of a new design that was the prototype for the Constellation class, of which Picard's U.S.S. Stargazer was a member.

The Defiant disappeared in 2268 near Tholian territory into what was believed to be a spatial interphase. This interphase had an adverse effect on humanoid neurophysiology and caused mass insanity among the crew prior to the disappearance of the ship. The Defiant was last seen shimmering, suspended between two dimensions, until it faded into interspace.

The Defiant emerged in the mirror universe during the 22nd century, where it was discovered that Tholians in that universe had created the interphasic rift by detonating a tri-cobalt warhead within the gravity well of a dead star, and had sent the distress call which had lured the Defiant to the area. The Tholians, soon after, took the ship to a drydock built into an asteroid-moon in orbit around a gas giant in the Vintaak system and began a salvage operation, but were unable to finish because the ship was stolen by Commander Jonathan Archer of the ISS Enterprise (NX-01).
After Enterprise was destroyed in a Tholian web, Archer (now on the Defiant) made a desperate attempt to escape by restoring shields and weapons. After the station tractored the Defiant, Tucker was able to restore those systems. The Defiant began firing at the Tholian station and disabled it. At first, the Defiant could only reach speeds at impulse because a Gorn stole one of the key components from the engines. The Gorn was killed by Archer and he was able to restore warp drive.
After arriving at the rendezvous point with the assault fleet, Archer found two Andorian battle cruisers, one Vulcan D'Kyr type cruiser, and one Tellarite ship were disabling the ISS Avenger. The Defiant destroyed all rebel ships except one Andorian cruiser which Archer intentionally allowed to escape.
Soval and T'Pol tried to sabotage Defiant with Phlox's help, inspired by the discovery that in the United Federation of Planets of the other universe, Vulcans and Denobulans were equal to humans. When Phlox disabled main power, the Defiant was defensless, and the Avenger, commandered by rebel species, began firing at her. Defiant recieved heavy damage until Tucker was able to restore main power. With power restored, Archer ordered the destruction of Avenger. The Defiant fired phasers and photon torpedos and destroyed Avenger's warp regulator. The second barrage of photon torpedos was targeted at their main reactor. The Avenger was destroyed with all hands. Soval and his mutinous crew died with him.
After destroying Avenger, Defiant arrived at Earth, now under the command of the self-proclaimed "Empress Hoshi Sato", having poisoned Archer and taken command of the ship. She ordered Admiral Gardner to surrender and standby for instructions, or she would open fire on Earth.

Perhaps the most famous spacecraft in the history of space exploration. Launched in 2245 from the San Francisco Yards orbiting Earth, the Enterprise was first commanded by Captain Robert April, then by Captain Christopher Pike.

Star Trek: The Original Series

We conjecture that Captain Pike commanded two five-year missions of the Enterprise before Kirk's tenure at the helm, and that Captain Robert April commanded a five-year mission before Pike. This is reasinably consistent with a commissioning date of 2245.

The Yorktown was scheduled to rendezvous with the Enterprise in 2268 to transfer critically neede vaccines for planet Theta VII. The Enterprise was unable to make the rendezvous with the Yorktown until the investigation and destruction of the dikironium cloud creature was complete.
TOS "Obsession"
Named for the American aircraft carrier that fought in the Pacific theater during World War II. Yorktown was the original name of Star Trek's spaceship, before the ship was called Enterprise, from Gene Roddenberry's first draft of the series outline written in 1964. Roddenberry repordly suggested that the second Starship Enterprise, NCC-1701-A, launched at the end of Star Trek IV, had previously been named the Yorktown, since it seems unlikely that Starfleet could have built an all-new ship so quickly. If this was the case, the Yorktown may have made it safely back to Earth and been repaired and reanamed, or perhaps there was a newer, replacement Yorktown alredy under construction at the time of the probe crisis.

From the Star Trek Starship Spotter
History:
One of Starfleet's most famous types of vehicle, the Constitution-class starships included the acclaimed original U.S.S. Enterprise. During the time of Captain James T. Kirk's celebrated first five-year mission of exploration, only 12 of these ships were in existence.
TOS "Tomorrow is Yesterday"
Constitution-calss ships used duotronic computers, based on designs developed by Dr. Richard Daystrom in 2243.
TOS "The Ultimate Computer"
The registry number of the Constitution (NCC-1700) if from one of Scotty's technical manual screens in TOS "Space Seed". Since the class ship has a 1700 number, it would seem only reasonable that the other ships of the class would have higher, possibly even sequential numbers. Unfortunately, the U.S.S. Constellation TOS "The Doomsday Machine", bore a much lower number, NCC-1017, (obviously because it was a simple rearrangment of the decal sheet from the AMT Enterprise model kit) and the Republic was designed as NCC-1371. These data points suggest the Constitution-class ships had registry numbers that not only varied widely in range, but also could not be sequential. Modelmaker Greg Jein (Through an amazingly complex and admittedly only barely logical means) managed to match up the varios Constitution ships with the starship status chart in Commodore Stone's office in Starbase 11, seen in TOS "Court Martial". Most of these registry numbers are from Greg's conjectural list, although several are from varios Starfleet charts and readouts in Star Trek VI. A few of the Constitution-class ships listed above are not from any episode or movie, but are from the original Star Trek production office's starship list in Stephen Whitfield's book, The Making of Star Trek.
